What is going on , is that UAC is blocking you from access. If you access the directory from a remote machine via the administrative share you will have access. If you want to have access, then the best option is to create another group that domain admins or local admins belong to and assign that group to the folder .
Why are there no permissions to access files?
In a lot of cases its just the UAC set to protect the pc so its a saftey issue were it stops you editing sensertive files until you say you want to run as admin. Some folders are system which is above admin so this stops a user interfering with windows files this is so malware even as admin cant screw up your pc.
